Trametinib (Mekinist) is a kinase inhibitor. As a representative targeted drug in the era of precision medicine, it has demonstrated favorable clinical value in the treatment of melanoma.

What Are the Indications for Trametinib (Mekinist)?

Indications

1. Trametinib is mainly indicated for patients with unresectable or metastatic melanoma who have been confirmed to harbor BRAF V600E or V600K gene mutations via an FDA-approved testing method.

2. Prior to medication administration, the mutation type must be clearly identified through genetic testing of tumor samples, which is a critical prerequisite for determining treatment benefits.

3. It is particularly important to note that this drug is explicitly not suitable for patient populations who have previously received BRAF inhibitor therapy, a restriction that has been clearly validated in clinical trials.

Dosage Forms, Specifications and Physical Characteristics of Trametinib (Mekinist)

Dosage Forms, Specifications and Physical Characteristics

1. 0.5 mg tablet: Yellow, modified oval-shaped, biconvex film-coated tablet, engraved with "GS" on one side and marked with "TFC" on the other.

2. 1 mg tablet: White, round, biconvex film-coated tablet, engraved with "GS" and marked with "LHE".

3. 2 mg tablet: Pink, round, biconvex film-coated tablet, marked with "GS" and "HMJ".

4. All tablets adopt a specific formulation: the tablet core contains mannitol, microcrystalline cellulose, hypromellose and other ingredients.

5. Different colorants are added to the coating layer according to specifications (iron oxide yellow for 0.5 mg tablets, iron oxide red for 2 mg tablets) to ensure the stability and bioavailability of the drug.

Storage Conditions for Trametinib (Mekinist)

Storage Conditions

1. Temperature control: The drug must be refrigerated and stored at a temperature of 2℃–8℃ (36℉–46℉).

2. Moisture protection: The desiccant placed in the original packaging shall be retained throughout the storage period and must not be discarded.

3. Light protection requirement: The drug should always be stored in its original bottle to avoid direct light exposure.

4. Prohibited practices: Freezing storage is strictly prohibited, and tablets should not be transferred to other pill boxes, so as to prevent reduced efficacy caused by moisture erosion.
