How well does NEXIUM(Esomeprazole) work?

The therapeutic effect of esomeprazole is mainly reflected in its powerful gastric acid inhibitory effect. As a proton pump inhibitor, esomeprazole can effectively inhibit the proton pump (H+/K+-ATPase) in gastric parietal cells, thereby blocking the final step of gastric acid secretion and significantly reducing gastric acid secretion. 

How well does NEXIUM(Esomeprazole) work?

1. Trial Design In this placebo-controlled, double-blind, crossover study, 16 healthy male volunteers took 20 mg of esomeprazole daily for one week. On day 7, in an acute experiment, subjects consumed 500 ml of beer within 5 minutes. Gastroesophageal reflux was then monitored for 3 hours by pH determination. Gastric emptying was measured by ultrasound, and blood concentrations of ethanol, cholecystokinin, and gastrin were determined. 

2. Setting of Trial Outcomes Compared with placebo (2.6%), esomeprazole treatment significantly reduced gastroesophageal reflux by 93% (0.2%, median time percentage with pH < 4) (p=0.001). 

3. Trial Results Esomeprazole significantly reduced gastroesophageal reflux in healthy volunteers after beer consumption. Although esomeprazole treatment did not prolong gastric emptying time of beer compared with placebo, this proton pump inhibitor significantly increased plasma gastrin concentration. When taking PPIs, moderate alcohol consumption does not exacerbate gastroesophageal reflux.
